Antibodies and antibody-based therapeutics, including monoclonal antibodies (mAbs), bispecific antibodies (bsAbs) and antibody-drug conjugations (ADCs), are powerful tools. While the diversity and complexity of tumors makes it difficult to develop effective drugs, the precision and engineering potential of antibodies makes them a powerful tool to direct the immune system to destroy cancer cells or deliver cytotoxic payloads with high specificity.

As a crucial type of cancer therapy, the development of antibody-based therapies must be de-risked and accelerated.

The Biologics workstream of the Oncode Accelerator program focuses on developing antibody-based immune therapy to harness the power of the immune system to tackle cancer. 

At the basis of antibody discovery, the Biologics Workstream uses a unique method to generate high affinity antibodies against hard-to-target membrane proteins, alongside novel approaches like single B-cell selection in combination with high-throughput sequencing . Leveraging these technologies and approaches will increase the success rate of finding the best antibody for downstream optimization and clinical validation.

Together with the Oncode Accelerator innovation Platforms (Patient Cohorts, Organoids and Artificial Intelligence) and program partners, the Biologics Workstream will increase the chance of clinical success by, for example, directly screening on patient material for efficacy. Resulting leads will be optimized for developability for downstream manufacturing.

By working closely with fellow experts in lead finding, lead optimization, and (pre-)GMP activities, the Biologics Workstream will truly accelerate preclinical antibody development.

Key to the success of the Oncode Accelerator program is the selection and execution of high-quality Demonstrator Projects which aim to both validate the Oncode Accelerator infrastructure while simultaneously addressing unmet medical needs. We invite international cancer researchers, from across academia and industry to apply for a Demonstrator Project and partner with us for preclinical development of promising assets.

By combining the key capabilities of our  Workstreams with the innovations from our Platforms we create a synergistic effect resulting in better therapies, tailored to individual patients or patient subgroups.

    Why should this be of interest?

    The Biologics Workstream offers broad coverage of antibody modalities in-house and through collaboration with world-class partners. Demonstrator Projects can enter the program at various stages of the drug development pipeline, from antibody generation to late-stage efficacy and GMP production. Our three innovation platforms (Patient Cohorts, Organoids and Artificial Intelligence) are integrated at several stages in order to de-risk and accelerate the development process.

    By increasing the initial number of antibodies to be screened, screening on clinically relevant material, and manufacturing more complex antibody constructs for clinical validation, the success rate of finding the best antibody for downstream optimization and clinical validation should be increased.  In addition, this should reduce the time and cost for therapy development, through early de-selection due to detection/predicting of liabilities, early process guidance for clinical manufacturing, direct toxicity analysis in phase 0 studies, and an optimized workflow design.

Patient Cohorts   Organoids  Artificial Intelligence

A Demonstrator Project can be executed at any stage of the Workstream pipeline and incorporates at least one of the innovative Platforms (Patient Cohorts, Organoids and/or Artificial Intelligence). The stages are designed to ensure that potential oncology therapies are thoroughly evaluated and de-risked before moving on to clinical trials. One stage of a pipeline will be one Demonstrator Project, eligible for up to 50% co-funding.
