Oncode Accelerator Supports International Collaboration to Develop Novel Neuroblastoma Therapy
On 9th April, Oncode Accelerator announced a new Demonstrator Project supporting innovative research from the Antibody Therapy Group at University Medical Center (UMC) Utrecht and JJP Biologics, a Polish biotechnology company. The international team will leverage Oncode Accelerator’s innovation Platforms – Artificial Intelligence (AI), Patient Cohorts, and Organoids – to develop a novel bispecific antibody therapy.
A New Strategy for Targeting a Broad Range of Cancers
GD2 is a protein that many different tumor types have in common – from neuroblastoma, a childhood cancer, to adult cancers like melanoma, glioblastoma, and bladder cancer. This commonality makes it an attractive target for cancer therapy. Monoclonal antibody therapy targeting GD2 has already been approved for treatment of neuroblastoma – but patients may experience severe side effects to this therapy.
In the latest Demonstrator Project supported by Oncode Accelerator, an international team from the Antibody Therapy Group at UMC Utrecht, led by Professor Jeanette Leusen, and JJP Biologics will work together to develop a next-generation antibody therapy with fewer side-effects. The team will use Oncode Accelerator’s innovation Platforms – well-defined Patient Cohorts, Organoids, and Artificial Intelligence (AI) – to advance their promising strategy for targeting GD2.
The Demonstrator Project focuses on developing a bispecific antibody. Natural antibodies are Y-shaped molecules in which each ‘hand’ at the top of the Y binds the same target protein – for instance, GD2. Bispecific antibodies differ in that each ‘hand’ can bind a different target protein. The new antibody design aims to combine targeting of tumor-associated GD2 with an improved ability to call the right immune cells into action. This should kill tumor cells more efficiently. Simultaneously, the scientists will tweak and optimize different features of the antibody with the aim to prevent the side-effects associated with current standard-of-care treatments, such as neuropathic pain.

Leveraging Innovations to Fast-Track Research
Developing novel cancer therapies is a high-risk, expensive and time-consuming process, and new treatments often benefit only a subset of patients. Use of innovative technologies can increase the likelihood that promising ideas advance through preclinical development and ultimately deliver meaningful patient impact.

In this Demonstrator Project, Jeanette Leusen’s team will make use of a broad spectrum of the innovations that Oncode Accelerator offers, and will collaborate with fellow Oncode Accelerator program members Dr. Patrick Kemmeren and Dr. Judith Wienke (Princess Máxima Center for Pediatric Oncology). The team will leverage AI-enabled tools to optimize key features of the novel antibody, and the Patient Cohorts and Organoids Platforms will provide access to valuable patient samples and patient-derived organoids – mini organs that can be grown in the lab – to test whether the new bispecific antibody efficiently kills tumor cells. Using these innovations will help ensure that the therapy in development is well-positioned for future translation into the clinic.
As the seventh Demonstrator Project to launch, this project underlines the value of providing developers of novel therapeutics with access to Oncode Accelerator’s innovative oncology ecosystem.

Op 9 april kondigde Oncode Accelerator een nieuw Demonstrator Project aan ter ondersteuning van innovatief onderzoek van de Antibody Therapy Group van het Universitair Medisch Centrum (UMC) Utrecht en JJP Biologics, een Pools biotechnologiebedrijf. Het internationale team zal de innovatieplatforms van Oncode Accelerator inzetten – Kunstmatige Intelligentie (AI), Patiëntcohorten en Organoïden – om een nieuwe bispecifieke antilichaamtherapie te ontwikkelen.
Een nieuwe strategie om een breed scala aan kankers te beïnvloeden
GD2 is een eiwit dat voorkomt bij veel verschillende typen tumoren – van neuroblastoom, een vorm van kinderkanker, tot kankers die voorkomen bij volwassenen zoals melanoom, glioblastoom en blaaskanker. Daardoor is het GD2 eiwit een aantrekkelijk doelwit voor kankertherapie. Bestaande monoclonale antilichaamtherapie gericht tegen GD2 is eerder al goedgekeurd voor de behandeling van neuroblastoom, maar patiënten kunnen daarbij ernstige bijwerkingen ervaren.
In het nieuwste Demonstrator Project van Oncode Accelerator zal een internationaal team – bestaande uit de Antibody Therapy Group van UMC Utrecht, onder leiding van professor Jeanette Leusen, en het Poolse biotechbedrijf JJP Biologics – samenwerken aan de ontwikkeling van een nieuwe generatie antilichaamtherapie met minder bijwerkingen. Het team maakt gebruik van alle drie de innovatieplatforms (AI, Patiëntcohorten en Organoïden) van Oncode Accelerator om hun veelbelovende strategie voor het gericht aanpakken van GD2 verder te ontwikkelen.
Het Demonstrator Project richt zich op de ontwikkeling van een zogenaamd ‘bispecifiek’ antilichaam. Natuurlijke antilichamen zijn Y‑vormige moleculen waarbij elke ‘arm’ aan de bovenkant van de Y zich bindt aan hetzelfde doelwit-eiwit – bijvoorbeeld GD2. Bispecifieke antilichamen verschillen hierin doordat elke ‘arm’ zich aan een ander doelwit-eiwit kan binden. Het nieuwe ontwerp combineert het richten op het tumor-geassocieerde GD2 met een verbeterd vermogen om de juiste immuuncellen te activeren. Dit moet leiden tot een effectievere vernietiging van tumorcellen. Tegelijkertijd zullen de onderzoekers verschillende eigenschappen van het antilichaam aanpassen en optimaliseren om bijwerkingen die gepaard gaan met de huidige standaardbehandeling, zoals neuropathische pijn, te voorkomen.

Innovaties benutten om onderzoek te versnellen
De ontwikkeling van nieuwe kankertherapieën is risicovol, kostbaar en tijdrovend, en nieuwe behandelingen zijn vaak slechts effectief voor een handvol patiënten. Het gebruik van innovatieve technologieën kan de kans vergroten dat veelbelovende ideeën succesvol door het preklinische ontwikkeltraject gaan en uiteindelijk echte impact hebben voor patiënten.

In dit Demonstrator Project zal het team van Jeanette Leusen gebruikmaken van een breed scala aan innovaties die Oncode Accelerator biedt en samenwerken met andere programmadeelnemers van Oncode Accelerator, waaronder dr. Patrick Kemmeren en dr. Judith Wienke (beiden werkzaam binnen het Prinses Máxima Centrum voor Kinderoncologie). Het team zal AI-ondersteunde tools gebruiken voor de optimalisatie van essentiële kenmerken van het nieuwe antilichaam. De platforms voor Patiëntcohorten en Organoïden bieden toegang tot waardevolle patiëntmaterialen en organoïden (mini-organen) die vanuit patiëntmateriaal en tumoren in het laboratorium kunnen worden gekweekt, om te testen of het nieuwe bispecifieke antilichaam tumorcellen efficiënt kan doden. Door deze patiëntgerichte innovaties te benutten, wordt ervoor gezorgd dat de therapie goed gepositioneerd is voor toekomstige vertaling naar de klinische praktijk.
Met de start van het zevende Demonstrator Project wordt opnieuw de waarde zichtbaar van toegang tot het innovatieve oncologie-ecosysteem van Oncode Accelerator voor ontwikkelaars van nieuwe therapieën.
