Oncode Accelerator Supports UPyTher in Developing New Treatment for Hard-to-Treat Cancer

Oncode Accelerator and Dutch biotech start-up UPyTher have launched a collaborative Demonstrator Project to develop a new treatment approach for peritoneal metastases — a form of cancer that is particularly difficult to treat. The collaboration aims to make treatment safer, simpler, and more effective for patients who currently have very limited options.

From Unmet Clinical Need to Built-for-Purpose Therapy

Peritoneal metastases occur when cancer cells – typically from colorectal, ovarian, and gastric cancers – break off from the main tumor and spread into the abdomen. It is estimated that more than 1 million people are confronted each year worldwide, but treatment options remain limited and outcomes are often poor.

Peter-Paul Fransen, CTO of UPyTher, an Eindhoven based spin-off from the Technical University Eindhoven, explains: “Because peritoneal metastases are characterized by many small tumors in the abdominal cavity, it can be very hard to destroy all the spread-out cancer cells. Systemic therapies delivered through the bloodstream show limited efficacy against peritoneal metastases compared to metastases in other organs.” Local drug delivery directly to the abdominal cavity has long been proposed as a strategy to improve treatment efficacy for peritoneal metastases. Delivering drugs directly to the abdominal cavity should achieve higher local drug exposure while limiting what physicians call ‘systemic toxicity’ – side effects due to the systemic delivery of the drug. However, most existing cancer drugs weren’t designed for this approach, and are rapidly cleared from the peritoneal cavity. As a result, most patients have very few effective options.

By focusing in on this group of patients, UPyTher aims to develop new treatment strategies specifically suited to their needs. Their Demonstrator Project with Oncode Accelerator is the next step in changing the status quo for patients with peritoneal metastases.

Reaching Peritoneal Metastases Everywhere They Hide

UPyTher’s answer to this challenge in cancer care is their unique hydrogel: a biomaterial that can mixed with anti-cancer drugs, and then injected or sprayed into the abdominal cavity. The liquid spreads throughout the abdomen and then forms a thin gel layer, helping anti-cancer drugs reach the many small tumors, and stay in place long enough to destroy them.

Altogether, this approach can make treatment both safer and more effective. “In the abdominal cavity, peritoneal metastases can be everywhere,” Peter-Paul explains. “So, the therapy should also be everywhere and be effective for a longer period of time.”

The newly launched Demonstrator Project will further develop this innovation towards the clinic. Backed by funding from the Dutch National Growth Fund (NGF), Oncode Accelerator was established to innovate and accelerate the development of cancer therapies in the Netherlands and abroad. Within the Oncode Accelerator program, development of new cancer therapies is done through Demonstrator Projects, which empower cancer researchers to accelerate and de-risk the development of their new oncology therapies.

In their Demonstrator Project, UPyTher will access the world-class tumor biology expertise and representative organoid (lab grown mini-organ/tumor) models available within the Oncode Accelerator’s growing public-private research and development ecosystem. They will collaborate with researchers at UMC Utrecht to test new drug combinations using advanced lab-grown tumor models known as organoids. These models closely resemble real patient tumors and allow researchers to better predict how new treatments may perform. If successful, the new gel-based combination therapies could lead to better outcomes for patients with peritoneal metastases.

This Demonstrator Project exemplifies how innovation and collaboration can translate fundamental science towards real-world impact.

Peter-Paul Fransen elaborates: “Through Oncode Accelerator, we gain access to oncology expertise and advanced models, co-funding to help bring our tech towards the clinic, and visibility within the broader scientific and clinical community.” The Demonstrator Project will help UPyTher build a foundation for future investment alongside future collaborations.

Oncode Accelerator geeft UPyTher een boost in strijd tegen moeilijk te behandelen kanker

Oncode Accelerator en biotech startup UPyTher uit Eindhoven zijn een samenwerking in de vorm van een Demonstrator Project gestart om een nieuwe behandelmethode te ontwikkelen voor peritoneale metastasen — een vorm van kanker die bijzonder moeilijk te behandelen is. De samenwerking is bedoeld om behandeling veiliger, eenvoudiger en effectiever te maken voor patiënten die momenteel slechte vooruitzichten hebben.

Van klinische noodzaak naar innovatieve behandeling

Peritoneale metastasen ontstaan wanneer kankercellen – meestal afkomstig van darm-, eierstok- en maagkanker – zich losmaken van de hoofdtumor en zich verspreiden naar de buik. Er wordt geschat dat hier wereldwijd jaarlijks meer dan 1 miljoen mensen mee te maken krijgen, maar de behandelingsmogelijkheden blijven beperkt en de vooruitzichten zijn vaak slecht.

Peter-Paul Fransen, Chief Technology Officer van UPyTher, een spin-off van de Technische Universiteit Eindhoven, legt uit: "Omdat peritoneale metastasen worden gekenmerkt door veel kleine tumoren in de buikholte, kan het erg moeilijk zijn om alle verspreide kankercellen te vernietigen. Bestaande therapieën die via de bloedbaan worden toegediend, werken minder goed tegen peritoneale metastasen vergeleken met uitzaaiingen in andere organen." Medicijnen rechtstreeks in de buik toedienen kan helpen om de behandeling effectiever te maken. Zo worden tumoren in de buik langer blootgesteld aan het medicijn, terwijl wat artsen ‘systemische toxiciteit’ noemen – oftewel het ontstaan van mogelijke bijwerkingen – wordt beperkt.

De meeste bestaande kankermedicijnen zijn echter niet voor deze aanpak ontworpen en worden daarom snel uit de buikholte verwijderd. Daardoor zijn de opties voor de meeste patiënten met deze peritoneale metastasen op dit moment weinig effectief en blijven de vooruitzichten slecht.

Door zich op deze groep patiënten te richten, wil UPyTher nieuwe behandelstrategieën ontwikkelen die specifiek zijn afgestemd op hun behoeften. Het Demonstrator Project met Oncode Accelerator is de volgende stap in het veranderen van de status quo voor patiënten met peritoneale metastasen. 

Peritoneale metastasen bereiken op plekken waar ze zich verstoppen

UPyTher’s antwoord is hun unieke hydrogel: een biomateriaal dat vermengd kan worden met antikankermedicijnen en vervolgens in de buikholte kan worden geïnjecteerd of gesprayd. De vloeistof verspreidt zich door de buik en vormt vervolgens een dunne laag, waardoor antikankermedicijnen de vele kleine tumoren bereiken en lang genoeg op hun plaats blijven om ze te vernietigen.

Al met al kan deze aanpak de behandeling zowel veiliger als effectiever maken. "In de buikholte kunnen peritoneale metastases overal zijn," legt Peter-Paul uit. "dus de therapie zou ook overal en langer effectief moeten zijn."

In het nieuwe Demonstrator Project zal deze innovatie verder worden doorontwikkeld richting de kliniek. Met financiering van het Nationaal Groeifonds is Oncode Accelerator opgericht om innovatie en de ontwikkeling van kankertherapieën in Nederland en daarbuiten te versnellen. Binnen het Oncode Accelerator-programma wordt de ontwikkeling van nieuwe kankertherapieën uitgevoerd via Demonstrator Projecten, die onderzoekers in staat stellen de ontwikkeling van hun nieuwe kankertherapieën te versnellen en tegelijkertijd de risico’s op falen te verminderen. 

UPyTher krijgt via het Oncode Accelerator Demonstrator Project toegang tot tumorbiologie-expertise van wereldklasse en representatieve organoïden. Dit zijn geavanceerde tumormodellen die worden gekweekt in het lab. Deze modellen lijken sterk op daadwerkelijke patiënten tumoren en stellen onderzoekers in staat beter te voorspellen hoe nieuwe behandelingen zullen reageren. De organoïden worden ter beschikking gesteld voor onderzoek binnen het groeiende publiek-private ecosysteem van Oncode Accelerator. UPyTher zal samenwerken met onderzoekers van het UMC Utrecht, een van de coördinerende partners van Oncode Accelerator, om nieuwe combinaties van medicijnen met de hydrogel van UPyTher te testen op de organoïden modellen. Als de nieuwe aanpak succesvol blijkt, kunnen de nieuwe gel-gebaseerde combinatietherapieën uiteindelijk leiden tot betere uitkomsten voor patiënten met peritoneale metastasen.

Samenwerking en innovatie: de sleutel tot betere behandelingen

Dit Demonstrator Project laat zien hoe innovatie en samenwerking fundamenteel wetenschappelijk onderzoek kunnen vertalen naar impact in de echte wereld.

Peter-Paul Fransen licht toe: "Via Oncode Accelerator krijgen we toegang tot oncologische expertise, geavanceerde modellen en publieke financiering om onze technologie naar de kliniek te brengen. Dit geeft ons ook de mogelijkheid om meer bekendheid voor onze aanpak en technologie te creëren." Het Demonstrator Project zal UPyTher helpen een basis te leggen voor nieuwe investeringen en mogelijke toekomstige samenwerkingen.

"Ons doel is eenvoudig maar ambitieus," concludeert Peter-Paul. "We willen patiënten met peritoneale metastasen een echte kans geven - met therapieën die speciaal voor hen zijn ontworpen, in plaats van behandelingen die als bijzaak worden toegepast maar die niet effectief zijn."

Door wetenschappelijke disciplines zoals biomaterialen, tumorbiologie en geneesmiddelenontwikkeling samen te brengen, werken UPyTher, het UMC Utrecht en Oncode Accelerator samen om kanker overal waar het zich verbergt te bereiken — en daarmee de uitkomsten te verbeteren voor patiënten met momenteel slechte vooruitzichten.
